How to Reconstitute Peptides: A Step-by-Step Laboratory Guide
Lyophilized peptides are shipped as a freeze-dried powder because peptides are far more stable dry than in solution. Reconstitution — returning that powder to a usable liquid — is the single step where more research material is ruined than anywhere else in a protocol. This guide covers the standard laboratory procedure.
1. Choose the right diluent
For most research peptides the choice is between two options:
- Bacteriostatic water (sterile water + 0.9% benzyl alcohol) — the default for any protocol lasting more than a day or two. The preservative inhibits microbial growth across repeated withdrawals from the same vial, giving a working life of up to 30 days refrigerated.
- Sterile water for injection — appropriate only for single-use, same-day protocols. Without a preservative, a punctured vial should not be stored and reused.
A small number of peptides with poor water solubility may call for a drop of acetic acid or a DMSO co-solvent first; the product's technical sheet will say so explicitly. Every product we sell reconstitutes in plain bacteriostatic water unless its page notes otherwise.
2. Calculate your volumes before opening anything
Decide the final concentration you want, then work backwards. The math is one line:
Concentration (mg/mL) = peptide mass (mg) ÷ diluent volume (mL)
Adding 2mL of diluent to a 10mg vial yields 5mg/mL — so every 0.1mL drawn contains 0.5mg. Adding 1mL to the same vial yields 10mg/mL. Larger diluent volumes make small doses easier to measure accurately; the trade-off is that our 3mL vials hold at most 3mL. Write the concentration and reconstitution date on the vial immediately — future you will not remember.
3. Reconstitute with gentle technique
- Let the peptide vial reach room temperature before opening (condensation on cold glass wets the powder unevenly).
- Swab both vial septa with an alcohol prep pad and let them dry.
- Draw the calculated diluent volume with a sterile syringe.
- Insert the needle through the peptide vial's septum and direct the stream against the glass wall, never onto the powder cake. Peptide bonds are mechanically fragile; a direct jet can shear them.
- Let the diluent run down and pool. Swirl gently or roll the vial between your palms until fully dissolved. Never shake — foaming denatures peptides.
- Most peptides dissolve within a minute or two into a clear solution. Persistent cloudiness or visible particulates after ten minutes means the vial should not be used.
4. Store the solution correctly
Refrigerate reconstituted peptides at 2–8°C and use within the validated window — typically 30 days with bacteriostatic water. Do not freeze reconstituted solution: ice crystals damage peptide structure, and repeated freeze-thaw cycles are worse. Common mistakes to avoid
- Shaking instead of swirling — the most common cause of degraded material.
- Injecting diluent directly onto the powder cake.
- Using plain sterile water for a multi-week protocol.
- Storing reconstituted vials at room temperature or in a freezer.
- Failing to label concentration and date, then guessing later.
