Immunomodulatory peptides tune immune signalling rather than bluntly stimulating or suppressing it. This category includes thymosin alpha-1, KPV, and VIP — three molecules with very different mechanisms that converge on immune regulation.

Thymosin alpha-1 is the most clinically documented: a 28-amino-acid thymic peptide studied as a TLR2/TLR9 agonist that promotes dendritic-cell activation and T-cell maturation, with regulatory approvals in over 30 countries outside the US. KPV is the opposite extreme in size — the three-residue C-terminal fragment of alpha-MSH that retains the parent hormone's anti-inflammatory signalling, best known from rodent inflammatory-bowel-disease models and its unusual PepT1-mediated intracellular uptake. VIP rounds out the category as a native neuropeptide with documented suppression of pro-inflammatory cytokines and promotion of regulatory T-cell responses.
