TB-500 (Thymosin Beta-4) Research Overview: Mechanisms and Study Models
TB-500 is the research-market name for synthetic thymosin beta-4 (Tβ4) and its active fragments. Unlike many research peptides, Tβ4 is a naturally abundant human protein with a well-mapped mechanism — which makes the literature unusually coherent.
Mechanism: actin sequestration
Thymosin beta-4 is a 43-amino-acid peptide and the body's principal G-actin sequestering molecule — it binds monomeric actin and regulates its polymerization into filaments. Because actin dynamics drive cell shape, division and movement, Tβ4 sits directly upstream of cell migration. The short central fragment containing the LKKTETQ actin-binding motif reproduces much of this activity and is the basis of several synthetic versions sold for research.
What the studies have examined
- Dermal wound healing — the foundational work: topical and systemic Tβ4 accelerated wound closure, increased keratinocyte and endothelial migration, and improved collagen deposition in rodent models, including diabetic and aged animals.
- Corneal repair — Tβ4 promoted re-epithelialization in corneal injury models, work that progressed into human trials of Tβ4 eye drops (RGN-259) for dry eye and neurotrophic keratopathy — among the few thymosin programs to reach clinical stages.
- Cardiac models — after coronary ligation in mice, Tβ4 supported cardiomyocyte survival and epicardial progenitor activation; this remains an active preclinical area.
- Anti-fibrotic and anti-inflammatory effects — reduced scar formation and inflammatory infiltration reported across several organ injury models.
Where the evidence stops
Outside the ophthalmic programs, Tβ4 research remains preclinical. No injectable TB-500 product is approved for human or veterinary use, and sports anti-doping agencies explicitly prohibit it. Published animal results — however consistent — do not establish human efficacy or safety.
Laboratory handling
TB-500 reconstitutes cleanly in bacteriostatic water and follows standard storage practice: refrigerated, dark, used within the 30-day window. Its larger size versus short peptides makes gentle handling during reconstitution more important — swirl, never shake.
