Peptide Calculations for Laboratory Work: Concentrations, Dilutions and Molarity
Most peptide-work errors are not chemistry errors — they are arithmetic errors made at the bench. Four calculations cover essentially every protocol. Work through them once and keep this page as the reference.
1. Reconstitution concentration
The master formula, one line:
Concentration (mg/mL) = peptide mass (mg) ÷ diluent volume (mL)
A 10mg vial plus 2mL of bacteriostatic water = 5 mg/mL. The same vial plus 1mL = 10 mg/mL. Choose the volume before opening anything, based on what makes your target quantity an easy, accurately measurable draw — small quantities are easier to measure precisely from more dilute solutions.
2. Quantity per draw
Quantity (mg) = concentration (mg/mL) × volume drawn (mL)
From a 5 mg/mL solution, a 0.1mL draw contains 0.5mg (500mcg); a 0.05mL draw contains 0.25mg (250mcg). Two unit habits prevent the classic tenfold error: write mcg (micrograms) out fully rather than "µg" scrawled by hand, and remember 1mg = 1000mcg. On graduated syringes, confirm whether markings are in mL or hundredths of a mL before trusting them for volume math.
3. Serial dilutions for dose-response work
In-vitro protocols usually need a concentration series (e.g. 10, 1, 0.1, 0.01 µg/mL). Build it by serial 1:10 dilution: transfer 0.1mL of stock into 0.9mL of diluent, mix gently, repeat from the new tube. Always pipette from the freshly mixed tube, change tips between steps, and make each series fresh — dilute peptide solutions lose material to tube walls faster than concentrated stocks.
4. Mass to molarity (in-vitro work)
Cell-culture literature reports peptide concentrations in molar units, so converting is routine:
Molarity (µM) = concentration (µg/mL) ÷ molecular weight (kDa) × 1000 — or more simply: µM = (µg/mL) ÷ MW in kDa when working in mg/L.
Worked example: BPC-157 (MW ≈ 1.42 kDa) at 1 µg/mL is 1 ÷ 1419 × 1,000,000 ≈ 0.70 µM. Semaglutide (MW ≈ 4.11 kDa) at the same 1 µg/mL is only ≈ 0.24 µM — a reminder that equal masses of different peptides are very different molar doses. Molecular weights are on every certificate of analysis.

Label every vial with concentration and date the moment it's mixed (reconstitution guide), and stock enough bacteriostatic water to avoid improvising volumes mid-protocol. For research use only — not for human or veterinary use.
